Be sure to stop in Middleburg (on US 50). There is no other town like it in the United States.
 
There are a couple on US 17, US 50 and on US 340 - one of them is Shenandoah winery off of US 340. The other poster did a good job with the web site - there are a lot more vineyards on the otherside of the mountains than in the Shenandoah Valley itself. Leesburg is another quaint town to visit as well as the Oatlands historical area on US 15 South of Leesburg.

If you can drive a little more to the north, there is Lignanore Vineyard in Frederick County, MD and the Boordy vineyard on MD 146 in Baltimore County. Both will be showing off their fall foilage in the coming weeks.
