If it was a joke she would have made it clear before you guys parted ways. If she truly is your bestfriend then it shouldnt matter.
I have a really good friend who is a lesbian, it was weird to hear it at first and we all just thought that she was doing it for attention. After a while we realized that she actually was and I am completely fine with it. I have tons of guy friends who never hit on me and she is the same way. She talks to me about her girl problems and I talk about guy problems. Just stay open minded and help your friend, I am sure it is really hard for her to come out to you - she has to really trust you.
Why would it matter? If she is your BEST friend already it shouldn't make it a difference at all. Why would it make you freak out?
Just play it cool. There's nothing wrong with gay people. I have a lot of gay friends that I love dearly.
Don't freak out because maybe she was just kidding.
I seriously doubt she would tell you that as a joke.

Its best to make your stand on lesbians clear if you wish to avoid any awkward moments. Tell her to keep her les tendencies to herself if shes around you but that you still wish to be friends with you.
But do it subtly, with respect for her perspective in life and as you would to a friend.
My friends didn't believe me either.
My one friend, who didn't believe, approached me and asked me again. She had her serious face on.
