You're mounting the image? And it's still telling you to insert the DVD rom? Not sure, try getting a No-CD crack, put it over the original .exe, see if that helps. I'm not familiar with Alcohol 120% but I think it mounts images. Try mounting it with Daemon Tools, see if you get the same result...

Not sure, but if it's a GBA emulator, what about Virtualboy Advance or TGB Dual? I prefer VBA but I use TGB Dual for the built in link cable emulation sort of thing.
